We need each gene to be read by at a depth of 100x when it is expressed at a low level. The sample number for each batch could vary from one (1) to twenty-four (24). Whole Genome Sequencing Question: what is genome size and sequencing depth/coverage required? is a PCR-free method required? How many samples? Answer: The size varies from 12 Mbp to 20 Mbp. We need at least 100x coverage depth. For whole genome sequencing, we don’t need to perform any PCR. Sample number could vary from 1 to 12 for each batch. Amplicon Sequencing Question: What is sequencing depth required? Is this a sequencing-only project? If after library QC and cleanup is required, would end-user accept the additional $sample charge? How many samples? Answer: Coverage depth is around 100x to 6000x. We can accept reasonable library QC and cleanup charges if needed. A2: In the upcoming year (2026) the CBE Department is estimating to spend approximately $30,000-$50,000. Additional years after may fall into a range of $20,000-$30,000. The CBE department could submit all six types in a year (Plasmid, Amplicon, Genome, AAV, Microbiome, RNA-seq).
