    AI Summary

The U.S. Naval Medical Research Unit SOUTH seeks to procure reagents for library preparation and sequencing on MiSeq and MiniSeq platforms. The kits must support low DNA input and include necessary components for high-quality sequencing. Delivery is required to Callao, Peru, and all materials must maintain a cold chain. Vendors should be aware of the time-sensitive nature of these reagents.

    Description

    PERFORMANCE WORK STATEMENT

    1. SCOPE

The laboratory of the Virology Department of the U.S. Naval Medical Research Unit SOUTH (NAMRU SOUTH) is looking to purchase reagents to prepare libraries using human samples that later will be sequenced in the MiSeq and MiniSeq platforms developed by Illumina, as part of the projects of next-generation sequencing (NGS) that are being conducted at this command.

      1. BACKGROUND

The kits must be designed for library preparation and sequencing. The library preparation kit should include a variety of steps that include enzymatic fragmentation, size selection, end-repair, a-tail and ligation step in the same reaction. The process should allow including dual index at the 5 and 3 prime ends for later sample identification. Also, the library preparation kit should be designed to construct libraries with a low DNA input (~50ng) with master-mixed reagents ready to use. Similarly, the sequencing kit should allow us to run more than 150 sequencing cycles. Those cycles are needed to run paired-end libraries with a high-quality performance. We expect to have more than 2.0Gb per sequencing run with high Q30 values. Finally, the sequencing kit should include an internal control to validate the sequencing run.

    Applicable Standards

These reagents are very time-sensitive and have limited expiration dates, reason for which, vendor should inform final users of expiration dates as soon as they become available and request their approval prior to shipment.
